General description
噻吩甲酰胺基丙酸酯化合物可作为直接竞争性和高亲和力抑制剂，抑制dsRNA和TLR3的结合（Ki = 2.96 µM；dsRNA结合TLR3的Kd = 19 nM），同时其可选择性拮抗经刺激的TLR3信号转导，且不会影响TLR1/2、TLR2/6、TLR4和TLR7途径。在Poly（I：C）诱导的RAW 264.7巨噬细胞中，其显示出可有效抑制TNF-α和IL-1β（IC90 = 27 µM）和NO的产生（IC50 = 3.44 µM）。其表现出最小的细胞毒性（在RAW 264.7细胞中IC50 >>100 µM），并对一组12种代表性激酶和4种细胞色素酶的活性有较弱的抑制作用（体外分析）。
