General description
Pre-hexon-linking protein IIIa (UniProt: P12537; also known as Capsid vertex-specific component IIIa, CVSC, Protein IIIa, pIIIa) is encoded by the L1 gene in Adenovirus. Adenovirus 5 belongs to the Adenoviridae family of non-enveloped viruses that are characterized by linear, double-stranded DNA genomes. Their capsid is an icosahedron that contains at least 12 different viral proteins. Adenovirus 5 is commonly used for gene transfer experiments due to its ability to infect a wide group of different cell types. It can harbor large genes in its genome incorporated via homologous recombination techniques. Pre-hexon-linking protein IIIa is cleaved near the C-terminal region (residues 570-571) by the viral protease during virion maturation to form the mature hexon-linking protein IIIa protein. Maturation primes the viral particle for stepwise uncoating by facilitating penton release and by loosening the condensed genome and its attachment to the icosahedral shell. Protein IIIa that is expressed in the late phase of the viral replicative cycle serves as a structural component of the virion that acts as a cement protein on the capsid exterior and which mediates the interactions between the hexons, including the peripentonal hexons. It also interacts with packaging protein 3 and this interaction is reported to promote correct genome packaging. (Ref.: Charman, M., et al. (2023). Nature. 616(7956):332-338; Pérez-Berná, AJ., et al.
