Biochem/physiol Actions

This antiserum cannot distinguish between Glutathione S-transferase A1 and A2, which share more than 95% sequence homology (UniProt P13745 & P10648).

General description

Glutathione S-transferase (GST) A1 (EC:2.5.1.18; UniProt P13745; GST Ya, GST Ya1, GST A1, GST class-alpha member 1) and GST A2 (UniProt P10648; GT41A, GST Ya2, GST A2, GST class-alpha member 2) are encoded by the Gsta1 gene (Gene ID 14857) and the Gsta2 gene (Gene ID 14858), respectively, in murine species. Glutathione S-transferases constitute a family of metabolic isozymes that are involved in detoxification by catalyzing the conjugation of the reduced form of glutathione (GSH) to xenobiotic substrates. The GST family can be subdivided into the cytosolic, mitochondrial, and microsomal (a.k.a. MAPEG) proteins. Mammalian GSTs exist in either as homodimers or heterodimers. The individual monomers were historically characterized by their quaternary structure and size, Yf (Mr 24 500), Yk (25 kDa), Ya1 (25.5 kDa), Ya2 (25.5 kDa), Yn (26.5 kDa), Yb1 (27 kDa), Yb2 (27 kDa) and Yc (28.5 kDa). The “Y” designation refers to the “Y” fraction (as apposed to the “X and Z” fractions) they were found in during purification by Sephadex G75 chromatography. Murine Ya1 and Ya2 are different by only 11 out of their sequences (223 a.a./A1 or 223 a.a./A2). In addition, GST Ya1/Ya2 sequences are higly conserved among murine and rat species (>85% identity), while less sequence homology is found when comparing to the corresponding human proteins. Rat GST YaYa dimer was historically referred to as "Ligandin" or GST L, and old nomenclature also referred to human alpha classe GST A1-1, A1-2 and A2-2 as, respectively, B1B1, B1B2 and B2B2, where B stands for basic.
