蛋白质印迹:1:100-1:500,使用ECL。建议的稀释缓冲液是PBS,封闭缓冲液是含有5%牛奶的PBS。识别36 kDa单体、二聚体和三聚体(取决于样品制备)。煮沸诱导视紫红质的聚集。

用rho 1D4抗体标记视紫红质(约占视网膜感光细胞外节膜盘蛋白的85%)的分离的牛视网膜感光细胞外节膜盘的蛋白质印迹。 SDS凝胶上的视网膜感光细胞外节膜盘数量: (a)2.5 mg;(b)0.63 mg;(c)0.16 mg;(d)0.04 mg。在较高的蛋白质含量下,可以观察到视紫红质的二聚体、三聚体和四聚体以及单体。

免疫组织化学/细胞化学:1:100-1:500在多聚甲醛和戊二醛固定的冰冻组织切片上。优选的固定是在2-8°C下用多聚甲醛固定4小时。建议的透化方法是0.2% Triton X-100。

免疫沉淀

最佳工作稀释度必须由最终用户确定。
