General description
Genome polyprotein (UniProt: P14335) is encoded by the POLY gene (Gene ID: 912267) in West Nile virus. West Nile virus (WNV) is a mosquito-borne positive-sense RNA virus of the Flavivirus genus within the Flaviviridae family. WNV genome encodes for three structural domains known as C, prM, and E and seven non-structural proteins (NS1, NS2A, NS2B, NS3, NS4A, NS4B, and NS5). These are translated as a single polypeptide, which is subsequently cleaved by viral and cellular proteases. NS1 (aa 792-1143) is a glycoprotein that is involved in immune evasion, pathogenesis, and viral replication. It is secreted from infected cells and may vary its oligomeric state between monomers, dimers (the primary form) and hexamers. Once cleaved off the polyprotein, it is targeted to three destinations: the viral replication cycle, the plasma membrane, and the extracellular compartment. Intracellular NS1 functions as an essential cofactor for viral replication, and it localizes to WNV replication sites, whilst cell surface and secreted NS1 act as immunomodulators. NS1 is required for formation of the replication complex and recruitment of other non-structural proteins to the ER-derived membrane structures. It is reported to antagonize the complement function and inhibit signal transduction originating from Toll-like receptor 3. (Ref.: Martín-Acebes MA, et al. (2012). World J Virol. 1(2); 51-70).
