General description

Cullin-4A (UniProt Q13619; also known as CUL-4A) is encoded by the CUL4A gene (Gene ID 8451) in human. Polyubiquitination of proteins is mediated by ubiquitin-activating enzymes (E1s), ubiquitin-conjugating enzymes (E2s), and ubiquitin ligases (E3s). There exist RING finger-type (really interesting new gene), HECT-type (homologous to E6-AP C-terminus), and U-box type of E3 ligases. Cullin-4 belongs to the cullin family of hydrophobic proteins that confer substrate specificity to the multimeric Cullin-RING E3 ubiquitin ligase (CRL) complexes by acting as scaffold proteins. The human genome encodes 8 cullin proteins (CUL 1, 2, 3, 4A, 4B, 5, 7 and 9). The activity of CRLs is regulated by Cullin Neddylation via the reversible conjugation of a small ubiquitin-type protein known as Neural precursor cell expressed developmentally down-regulated protein 8 (Nedd8). CUL4A interacts with the substrate-binding adaptor DDB1 (DNA damage binding protein 1) via its arc-shaped helical N-terminal domain, and binds the small RING finger protein ROC1 (regulator of cullins 1) via its globular C-terminal domain. Despite the 83% sequence identity and seemingly functional redundancy between CUL4A and CUL4B, much more focus is placed on CUL4A in oncogenesis research.
