General description

Receptor-interacting serine/threonine-protein kinase 2 (EC 2.7.11.1; EC 2.7.10.2; UniProt O43353; also known as Receptor-interacting protein 2, CARD-carrying kinase, CARD-containing IL-1 beta ICE-kinase, CARD-containing interleukin-1 beta-converting enzyme-associated kinase, Growth-inhibiting gene 30, Receptor-interacting protein-like interacting caspase-like apoptosis regulatory protein kinase, RIP-2, RIP-like interacting CLARP kinase, Tyrosine-protein kinase RIPK2) is encoded by the RIPK2 (also known as CARD3, CARDIAK, CCK, GIG30, RICK, RIP2) gene (Gene ID 8767) in human. RIPK2 is a dual-specificity kinase that exhibits both serine/threonine as well as tyrosine kinase activities. RIPK2 participates in transducing cellular inflammatory response by propagating signaling downstream of the intracellular peptidoglycan sensors NOD1 and NOD2.

A major focus of breast cancer research is to understand the mechanisms responsible for disease progression and drug resistance. Toward that end, it has been found that approximately two thirds of all human breast carcinomas overexpress the Estrogen Receptor α (ERα) protein and it remains the primary pharmacological target for endocrine therapy1,2. The normal cellular function of ERα is as a transcription factor that mediates a wide variety of physiological processes, many of which are dependent upon phosphorylation of the receptor at specific amino acid residues3,4. Indeed, ERα is known to be phosphorylated at a multitude of different sites, yet how these all correlate to disease remains unclear5. Here, we interrogated multiple sites of ERα for phosphorylation status by screening an extensive panel of different breast cancer patient samples and other non-breast cancer tissue microarray (TMA) slide samples to determine their relevance to disease.
