General description

Antigen-presenting glycoprotein CD1d1 (UniProt P11609; also known as CD1d.1, T-cell surface glycoprotein CD1d1) is encoded by the Cd1d1 (also known as Cd1.1, Ly-38, AI747460) gene (Gene ID 12479) in murine species. CD1d is a nonpolymorphic MHC class I-like molecule, which presents nonpeptide ligands (e.g. glycolipids) to Type 1 or invariant natural killer T (iNKT) cells. iNKT cells differ from conventional T cells in that they recognize glycolipid antigens presented by the nonclassical β2-microglobulin (β2m)-associated CD1d. Recognition of CD1d-presented glycolipids by iNKT cells elicits the rapid secretion of both T helper type 1- (TH1-) and T helper type 2- (TH2-) associated cytokines without the need for priming. Murine CD1d is initially produced with a signal peptide sequence (a.a. 1-21), the removal of which yields the mature CD1d with a large extracellular region (a.a. 22-305), a transmembrane domain (a.a. 306-326), and a short cytoplasmic tail (a.a. 327-336).

Flow Cytometry Analysis: A representative lot detected α-galactosylceramide-/αGC-complexed CD1d on murine MH-S alveolar macrophages, LA-4 lung adenoma, and JAWSII bone marrow-derived dendritic cells (Rizvi, Z.A., et al. (2015). Toxicol. In Vitro. 29(6);1275-82.
Flow Cytometry Analysis: A representative lot detected αGC C20:2- and αGC26:0-complexed CD1d on the subpopulation of splenocytes with high CD11c-expression from mice injected with the respective α-GalCer analogs (Arora, P., et al. (2014). Immunity. 40(1):105-116).
Immunoprecipitation Analysis: A representative lot immunoprecipitated monoglycosylceramides-complexed mCD1d from rat RBL-Cd1d cells stably expressing murine CD1d (Kain, L., et al. (2014). Immunity. 41(4):543-554).
Immunocytochemistry Analysis: A representative lot detected α-GalCer-complexed murine CD1d on the surface of rat RBL-Cd1d cells stably expressing murine CD1d by fluorescent immunocytochemistry (Kain, L., et al. (2014). Immunity. 41(4):543-554).
Immunohistochemistry Analysis: A representative lot detected αGC C20:2- and αGC26:0-complexed CD1d on splenocytes in frozen spleen sections from mice injected with the respective α-GalCer analogs (Arora, P., et al. (2014). Immunity. 40(1):105-116).
ELISA Analysis: A representative lot selectively reacted with KRN7000-/α-GalCer-, OCH-, or C20:2-complexed mCD1d, but not glycolipid-free mCD1d. Clone L363 does not react with iGb3-complexed mCD1d, KRN7000-complexed hCD1b or KRN7000-complexed hCD1d (Yu, K.O., et al. (2007). J. Immunol. Methods. 323(1):11-23).

This Anti-CD1d/KRN7000 Antibody, clone L363 is validated for use in Flow Cytometry, Immunoprecipitation, Immunocytochemistry, Immunohistochemistry and ELISA for the detection of CD1d/KRN7000.

Biochem/physiol Actions

Clone L363 selectively reacts with KRN7000-/α-GalCer-, OCH-, or C20:2- complexed mCD1d, but not glycolipid-free mCD1d. Clone L363 does not react with iGb3-complexed mCD1d, KRN7000-complexed hCD1b or KRN7000-complexed hCD1d (Yu, K.O., et al. (2007). J. Immunol. Methods. 323(1):11-23).

Preparation Note

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.

Analysis Note

Evaluated by Flow Cytometry in alpha-GalCer glycolipid-treated RMA-S/mCD1d (WT) cells.

Flow Cytometry Analysis: 0.1 µg of this antibody detected CD1d/KRN7000 in alpha-GalCer glycolipid-treated RMA-S/mCD1d (WT) cells.
