General description

The murine gene Nup98 (Gene ID 269966; also known as AI849286, 4732457F17) encodes the 1816-amino acid nuclear pore complex protein Nup98-Nup96 (UniProt Q6PFD9), which is posttranslationally autocleaved between aa 800 and 801 to form Nup98 (aa 1-800) and Nup96 (aa 801-1816). The nuclear pore complex (NPC) acts as a passageway for nucleocytoplasmic transport. The core structure of the NPC is maintained by the Nup93-205 complex sandwiched between the supporting structures of the Nup107-160 complex on the nuclear and cytoplasmic faces of the pore. Nup96 is a component of the Nup107-160 complex, the largest subunit of the NPC. Nup96 knockout is reported to be fatal in mice and Nup96(+/-) mice with low levels of Nup96 display impaired immune response and are highly susceptible to viral infection.

A major focus of breast cancer research is to understand the mechanisms responsible for disease progression and drug resistance. Toward that end, it has been found that approximately two thirds of all human breast carcinomas overexpress the Estrogen Receptor α (ERα) protein and it remains the primary pharmacological target for endocrine therapy1,2. The normal cellular function of ERα is as a transcription factor that mediates a wide variety of physiological processes, many of which are dependent upon phosphorylation of the receptor at specific amino acid residues3,4. Indeed, ERα is known to be phosphorylated at a multitude of different sites, yet how these all correlate to disease remains unclear5. Here, we interrogated multiple sites of ERα for phosphorylation status by screening an extensive panel of different breast cancer patient samples and other non-breast cancer tissue microarray (TMA) slide samples to determine their relevance to disease.
