General description

DnaJ homolog subfamily B member 1 (UniProt Q3MI00) is encoded by the DNAJB1 gene (Gene ID 538426) in bovine species. Heat shock proteins (Hsps) are classified into six major families based on their relative molecular mass, Hsp110/hspH, Hsp90/HspC, Hsp70/HspA, Hsp60/HspD, Hsp40/DnaJ, and small heat shock proteins sHsp/HspB. Hsp40/DnaJB1 belongs to the Hsp40/DnaJ subfamily of heat shock proteins. Hsp40 acts as a co-chaperone with Hsp70 to facilitate protein folding, transport, and degradation. MK5 is reported to stimulate the ATP hydrolyse activity of Hsp40/Hsp70 complex by phosphorylating Hsp40/DnaJB1 C-terminal serine residues Ser149, Ser151, Ser171, resulting in an enhancement of Hsp40/DnaJB1-dependent suppression of heat shock factor 1-driven transcription. DnaJB1 is also shown to interact with MDM2 and prevent MDM2-mediated ubiquitination and proteasomal degradation of p53. DnaJB1 depletion promotes cancer cell growth as a result of p53 pathway inactivation.
~38 kDa observed

Purified bovine DNAJB1.

Application

Immunocytochemistry Analysis: 1-5 µg/mL from a representative lot detected heat shock-induced DNAJB1 upreguation in C6 rat glioma, B16 mouse melanoma, and K-562 human erythroleukemia cells (Courtesy of Dr. Irina V. Guzhova and Dr. Boris Margulis, Russian Academy of Sciences).
Flow Cytometry Analysis: 1-5 µg/mL from a representative lot detected heat shock-induced DNAJB1 upreguation in C6 rat glioma, B16 mouse melanoma, and K-562 human erythroleukemia cells (Courtesy of Dr. Irina V. Guzhova and Dr. Boris Margulis, Russian Academy of Sciences).
Western Blotting Analysis: 0.05-0.5 µg/mL from a representative lot detected heat shock-induced DNAJB1 upreguation in C6 rat glioma, B16 mouse melanoma, and K-562 human erythroleukemia cells (Courtesy of Dr. Irina V. Guzhova and Dr. Boris Margulis, Russian Academy of Sciences).
This Anti-DNAJB1/Hdj1 Antibody, clone J25 is validated for use in Western Blotting, Immunocytochemistry and Flow Cytometry for the detection of DNAJB1/Hdj1.

"Mods – modifications – small alterations can have a big impact on form and function. In the motorsports world, stock vehicles are modified to enhance their performance. Modifications, or mods, to the engine, drive train, intake and exhaust components add up to provide phenomenal performance gains that can be measured as horsepower and torque increases, which yield a competitive advantage, and result in reduced run times. In biology, proteins undergo modifications that alter their function. Some proteins require the modifications in order to perform their function effectively, whether it’s a pro-protein that is cleaved to produce an active enzyme, or a protein that is phosphorylated to facilitate a signaling process. Other proteins, such as histones, undergo modifications that regulate gene expression and alter cellular function. There are several post translational modifications such as acetylation, methylation, phosphorylation, and ubiquitination that impact protein function and activity. As a result, the analysis of proteins and their post-translational modifications are particularly important for the study of normal and disease-associated processes. New antibodies to detect phospho Histidines are now available from EMD Millipore."
