General description
Zinc finger and BTB domain-containing protein 16 (UniProt: Q05516; also known as Promyelocytic leukemia zinc finger protein, Zinc finger protein 145, Zinc finger protein PLZF) is encoded by the ZBTB16 (also known as PLZF, ZNF145) gene (Gene ID: 7704) in human. PLZF is a member of the krueppel C2H2-type zinc-finger protein family with nine C2H2-tpe zinc-finger domains. It serves as a transcriptional repressor and this repression may be mediated through recruitment of histone deacetylases to target promoters. It affects diverse signaling pathways, including growth-regulatory and differentiation pathways and is a key regulator of myeloid development. Its expression is observed in early, but in differentiated hematopoietic cells. It is expressed in bone marrow, early myeloid cells, and peripheral blood mononuclear cells. At lower level it is also detected in the ovary, kidney, and lung tissue. PLZF is reported to be essential for development of the characteristic innate/effector functions of natural killer T (NKT) cells. It is highly expressed during the differentiation of NKT cells and is essential for the acquisition of effector/memory innate-like functions. PLZF-deficient NKT cells lose their characteristic memory phenotype, and their function is significantly impaired. These cells lose their unique ability to co-secrete IL-4 and IFN- upon ex vivo stimulation. (Ref.: Kolesnichenko, M., and Vogt, PK. (2011). Cell Cycle.10(5); 771-775; Kovalovsky, D., et al. (2010).
