General description
Aiolos is a zinc finger DNA binding transcription factor important in lymphocyte differentiation and development. Aiolos plays an essential role in the regulation of B-cell differentiation including, proliferation and maturation to plasma cells. Aiolos also works as a cofactor in gene suppression via its interaction with the transcription factor FOXP3 in regulatory T cells as well, and Aiolos is involved in various chromatin functions too through its interaction with histone deacetylase complexes containing HDAC1, MTA2 and SIN3A. Aiolos is localized in the nucleus primarily but can be found in the cytoplasm depending upon the cellular situation. Aiolos is expressed in leukocytes and found in high concentration in spleen and thymus and has multiple isoforms. Activity is regulated via phosphorylation.

Hotspot mutation of IKZF3 (IKZF3-L162R) has been identified as a putative driver of chronic lymphocytic leukemia (CLL), but its function remains unknown. Here, we demonstrate its driving role in CLL through a B cell-restricted conditional knockin mouse model. Mutant Ikzf3
