General description
DNA-binding protein (UniProt: P03265; also known as DBP, Early 2A protein, Early E2A DNA-binding protein) is encoded by the DBP gene (Gene ID: 1460850) in human Adenovirus 5. Adenovirus 5. Adenovirus 5 belongs to the Adenoviridae family of non-enveloped viruses that are characterized by linear, double-stranded DNA genomes. Their capsid is an icosahedron that contains at least 12 different viral proteins. Adenovirus 5 is commonly used for gene transfer experiments due to its ability to infect a wide group of different cell types. It can harbor large genes in its genome incorporated via homologous recombination techniques. DNA-binding protein (DBP) is a viral component absolutely required for viral DNA replication. It plays a role in the elongation phase of viral strand displacement replication by unwinding the template in an ATP-independent fashion and thus employing its capacity to form multimers. It also enhances the rate of initiation. DBP influences the transcription of viral genes by interacting with viral promoters and enhancers. The expression of DBP is tightly regulated during the viral life cycle and is expressed early in the infection process, following the expression of immediate early genes. DNA-binding protein is mainly localized in the nucleus and which is consistent with its role in viral DNA replication and transcription regulation. DBP contains a short binding region (C-terminal arm; aa 513-529) that bridges DBP molecules together and creates a chain. It homomultimerizes on viral ssDNA bound to pTP and forms a initiation complex with viral polymerase, pTP and hosts NFIA and POU2F1/OCT1. DBP can interfere with host immune responses, aiding in immune evasion and contributing to the persistence of the virus in the host. Studies show that in adenovirus-infected cells in which gzmB and downstream apoptosis pathways are inhibited, an orphan granzyme gzmH directly cleaves the adenovirus DNA-binding protein (DBP) and induces an important decay in viral DNA replication.
