血管紧张素-转化酶-2(ACE2)通过水解血管紧张素1生成血管紧张素(1-9)。血管紧张素(1-9)是血管紧张素(1-7)的前体。ACE2也可能通过血管紧张素II的转化直接产生血管紧张素(1-7),血管紧张素II参与血管收缩。(2)血管紧张素(1-7)在肾素-血管紧张素系统(RAS)信号通路中起重要作用,并参与缓激肽诱导的血管舒张。因此,ACE2拮抗其同系物ACE的作用。ACE2对赖诺普利、卡托普利等常用抑制剂不敏感。由于血管紧张素II的过度活跃,该基因在心力衰竭中上调。研究ACE2对高血压和心力衰竭的潜在治疗价值。它与病毒包膜蛋白刺突(S)蛋白结合,并作为人类细胞中严重急性呼吸综合征冠状病毒(SARS-CoV)和SARS-CoV-2的主要进入受体。因此,它被认为是COVID-19的潜在治疗靶标。ACE2表现出对肺损伤的保护作用。

General description
血管紧张素转化酶2(ACE2)是一种跨膜蛋白,属于金属肽酶(M2)家族。是一种膜相关羧肽酶,在心脏、肾脏、睾丸、胃肠系统的血管内皮细胞和肺的上皮细胞中表达。ACE2由N末端肽酶结构域(PD)和C末端Collectrin样结构域(CLD)组成。它还包含与ACE相似的单个催化结构域。ACE2基因被定位到人类染色体位置Xp22.2。ACE2是结构同系物ACE。
观测分子量〜62 kDa。由于选择性剪接,单向转运描述了约92 kDa(亚型1)和约64 kDa(亚型2)的2个亚型。
