General description
E3 ubiquitin-protein ligase RNF43 (UniProt: Q68DV7; also known as RING finger protein 43, RING-type E3 ubiquitin transferase RNF43) is encoded by the RNF43 gene (Gene ID: 54894) in human. RNF43 plays a critical role in negatively regulating Wnt signaling, a pathway essential for cell proliferation, differentiation, stem cell maintenance, embryonic development and tumorigenesis. RNF43 contains an N-terminal extracellular domain (aa 24-197), a transmembrane domain (aa 198-218), and a large cytoplasmic region (aa 219-783) that contains an atypical RING-type zinc finger (aa 272-313) and a pro-rich (aa 569-760) region. The RING finger domain allows it to facilitate the transfer of ubiquitin from an E2 enzyme to target proteins. This activity is central to its role in modulating the Wnt signaling pathway, as RNF43 ubiquitinates Frizzled receptors, leading to their degradation and thus to downregulation of the Wnt signaling. Phosphorylation of conserved serines also plays a role in regulating the function and oncogenic potential of RNF43. Missense mutations, or truncations of RNF43 have been implicated in several cancers, including colorectal, gastric, and pancreatic cancers. In addition, RNF43 mutations are associated with cystic fibrosis-related intestinal cancer and serrated polyposis syndrome, highlighting its importance in maintaining epithelial integrity.
