General description
Inactive rhomboid protein 2 (UniProt: Q80WQ6; also known as iRhom2, Rhomboid family member 2, Rhomboid veinlet-like protein 6, Rhomboid-related protein) is encoded by the Rhbdf2 (also known as Irhom2, Rhbdl6, Rhor) in murine species. RHBDF2/iRhom2, a multi-pass membrane protein with seven transmembrane domains, is predominantly localized in the endoplasmic reticulum (ER) membrane. It is reported to be crucial for the maturation and function of ADAM17 in myeloid cells. Hence, mice lacking this protein are protected from ADAM17- and TNF -dependent septic shock and inflammatory arthritis. ADAM17, a sheddase of the EGF receptor ligands and TNF , is shown to be involved in cell survival, proliferation, migration, and inflammation. However, it does not exhibit any protease activity on its own. RHBDF2 is involved in negative regulation of protein secretion and regulation of EGF receptor signaling pathway. By interacting with EGF, it may retain EGF in the endoplasmic reticulum and regulate its degradation through the ER-associated degradation (ERAD). (Ref.: Weskamp, G., et al. (2020). J. Biol. Lipopolysaccharide (LPS; 100 ng/mL; 24 h)-treated wild-type macrophage fractions from mouse bone marrow and RHBDF2 knockouts were probed with Cat. No. MABS2314, Anti-RHBDF2, clone RHF2B 11H7 (1:500 Dilution). Proteins were visualized using a Goat Anti-Rat IgG2a secondary antibody conjugated to HRP and a chemiluminescence detection system.
