Gene Information
human ... CEP250(11190)
General description
Centrosome-associated protein CEP250 (UniProt: Q9BV73; also known as 250 kDa centrosomal protein, Cep250, Centrosomal NEK2-associated protein 1, C-Nap1, Centrosomal protein 2) is encoded by the CEP250 (also known as CEP2, CNAP1) gene (Gene ID: 11190) inn human. Cep250 is a component of the core centrosome that is ubiquitously expressed. It can exist as a monomer and homodimer. It specifically associates with the proximal ends of both mother and daughter centrioles during interphase and in mitotic cells it dissociates from the mitotic spindle poles. At the end of cell division, it is shown to re-accumulate at centrosomes. Cep250 is differentially phosphorylated during cell cycle and phosphorylation status regulates its association/dissociation from centrosome. During M phase its C-terminal part is shown to be phosphorylated by NEK2. Two isoforms of Cep250 have been described that are produced by alternative splicing. Loss of Cep250-mediated centriole cohesion leads to an altered cell migration phenotype.
~280 kDa observed; 281.14 kDa calculated. Uncharacterized bands may be observed in some lysate(s).
Immunogen
Recombinant fragment corresponding to 455 amino acids from the C-terminal region of human Centrosome-associated protein CEP250.
Application

Centrosome amplification is a hallmark of cancer and PLK4 is one of the responsible factors for cancer associated centrosome amplification. Increased PLK4 levels was also shown to contribute to generation of cells with centriole amplification in mammalian tissues as olfactory
