General description
Rab5A is a membrane associated Ras related GTPase that is required for endosome fusion with the plasma membrane. Rab5A also contributes to filopodia extension and other cellular cytoplasmic extensions. Rab5A plays important roles in cellular differentiation, neurogenesis, exocytosis and protein transport. Rab proteins are a large family of GTP-binding proteins that regulate cellular membrane traffic and organelle identity. When membrane bound, each Rab has a very specific cellular location and recognizes distinct subsets of intracellular membranes, which allows Rab proteins to act as as key cellular regulators, determining the recruitment of downstream effectors to the correct membrane at the correct time. Recent research indicates that the localization of Rab proteins happens by sensing the stored curvature elastic energy in the membrane and the specific RabGEFs (Rab-activating guanosine diphosphate/guanosine triphosphate exchange factors) that associate with a particular Rab. Rab5A associates with Rabex-5 and this association provides the determinants for Rab5A specific membrane targets.
