General description
Purified mouse monoclonal antibody generated by immunizing BALB/c mice with the specified immunogen and fusing splenocytes with SP2/0-AG14 plasmacytoma cells (see application references). Recognizes the ~24-26 kDa bcl-2 protein.
Recognizes the ~24-26 kDa Bcl-2 protein in MCF-7 cells, tonsil, and lymph node tissue.
The bcl-2 proto-oncogene is encoded by a gene 230 kb in size that gives rise to a ~24-26 kDa protein which has been localized to the mitochondrial membrane. Although some data exists for implicating bcl-2 as a novel GTP binding protein, the major function of bcl-2 appears to be an inhibitor of apoptosis or "programmed cell death". It has now been well documented that bcl-2 becomes deregulated in tumor cells as a result of translocation into the immunoglobulin heavy-chain locus and is therefore constitutively activated in B cell malignancies (i.e. follicular lymphoma and chronic lymphocytic leukemia). In addition, bcl-2 has also been shown to protect mammalian cells from sodium azide, colchicine, DNA, RNA, and protein-synthesis inhibitors, as well as steroids, heat shock and irradiation.
Immunogen
a synthetic peptide (GAAPAPGIFSSQPG) corresponding to amino acids 41-54 of human Bcl-2
Application
Frozen Sections (1-5 μg/ml)
Immunoblotting (1-5 μg/ml)
Paraffin Sections (1-10 μg/ml, see comments)
Immunoblotting (1-5 μg/ml)
Paraffin Sections (1-10 μg/ml, see comments)
Physical form
In 50 mM sodium phosphate buffer, 0.2% gelatin, pH 7.5.
Analysis Note
Positive Control
MCF7 cells or tonsil or lymph node tissue
MCF7 cells or tonsil or lymph node tissue
Other Notes
Generally, no pre-treatment of paraffin sections is needed, but treating with heat may improve staining in some tissues. Antibody should be titrated for optimal results in individual sample types.
