General description
Understanding human neurodevelopment and the underlying biology remains a complex issue. Stem cell biology improvements have introduced new tools and more useful model systems. Human neural stem cells (hNSCs) and neural progenitor cells (NPCs) have provided a robust system in which to investigate the complex neurobiology questions in modern science. NPCs have the capability to differentiate into different neural cell types such as astrocytes and oligodendrocytes. They are useful in screening new, potentially therapeutic drugs in a highly cost-efficient and high-throughput manner due to the reliable and robust source of cells. NPCs can also be easily manipulated, making them an ideal platform to use.1 The ReNcell® Human Neural Progenitor group of cell lines are conditionally immortalized human neural progenitor cells which appropriately retain characteristics of their original tissue sites. Furthermore, the cells are genetically modified with a chemical switch (conditional immortalization) to give a greater control of proliferation and differentiation. While these cells are grown in their progenitor state, they can be terminally differentiated into neurons with distinct characteristics reflective of their original brain regions. This makes the ReNcell® hNPCs a strong source of distinct neural cells in which to further investigate neurobiology. Each line was conditionally immortalized using retroviral integration of a single copy of the c-mycERTAM construct.1 This allows the cells to retain proliferative properties when grown with the necessary growth factors and 4-hydroxytamoxifen (4-OHT). With changes in media formulation, the cells start to differentiate into mature neurons characteristic of their original tissue region. The ReNSC5 cell line is originally sourced from human ventral mesencephalon. The ReNcell® hNPC lines have a high number of potential applications in which they can be utilized such as disease modelling, drug discovery, neural cytotoxicity testing, and target validation.
