地高辛是一种半抗原，可用于核酸标记和检测系统。

应用

使用抗地高辛抗体检测地高辛标记的化合物，所用方法：
  • ELISA
  • 免疫组织细胞化学
  • 原位杂交
  • 蛋白质印迹
抗地高辛配基已用于DNA束缚。其已被用于将DNA分子附着到流动池的玻璃表面上。

生化/生理作用

存在Na+、Mg2+和ATP的情况下，地高辛可抑制钠离子泵。
来自绵羊的多克隆抗体对地高辛配基和地高辛具有特异性，并且与其他类固醇（如人类雌激素和雄激素）没有交叉反应性。

制备说明

工作浓度：抗体的工作浓度取决于应用类型和底物。以下浓度可作为指导：
  • ELISA：适于包膜：2至4μg/ml
  • 免疫组织细胞化学：0.5至2μg/ml
  • 原位 杂交：0.2至0.4μg/ ml
  • 蛋白质印迹：0.5至2μg/ml

工作溶液：用于包膜应用：PBS（磷酸盐缓冲盐水），pH 7.4
加入1ml PBS至终浓度为200μg/ml。
用地高辛免疫后，通过离子交换层析纯化绵羊IgG，并通过免疫吸附分离特异性IgG。

与其他类固醇无交叉反应，如人体雌激素（例如雌二醇）或雄激素（例如睾丸激素）。
