特异性

胶原酶降解天然胶原蛋白。中性蛋白酶® 是一种非特异性的金属蛋白酶。

应用

胶原酶/中性蛋白酶® 用于从多种不同的组织和器官中制备细胞。

组分

EC 3.4.24.4 & 3.4.24.3

单位定义

一个单位是指，在1分钟内在反应条件下释放出1 μmol酪氨酸对应的Folin-阳性氨基酸和多肽所需的酶活性。

制备说明

激活剂：Ca2+

工作浓度：约为1mg/mL（胶原酶：0.1U/mL，中性蛋白酶：0.8U/mL

酶活性：
胶原酶：0.1 Wünsch U/mg冻干粉（+ 25°C，4-苯基-偶氮苄基-氧基羰基-Pro-Leu-Gly-Pro-D-Arg作为底物）
中性蛋白酶®：0.8 Wünsch U/mg冻干粉（+ 37°C，酪蛋白为底物，pH 7.5）
污染蛋白水解活性：<0.1U/mg 冻干粉 (血红蛋白作为底物)

工作溶液：工作溶液制备 将酶溶于水中，使其终浓度为100mg/mL。用PBS(不含Ca2+/Mg2+)稀释至终浓度为1mg/mL，并采用无菌0.2μm 过滤膜过滤。
吴军溶液的酶浓度：胶原酶，0.1U/mL PBS; 中性蛋白酶®, 0.8U/mL PBS。

抑制剂：
胶原酶抑制剂：EDTA, EGTA
血清不能抑制胶原酶/中性蛋白酶。

保存条件（工作溶液）：-15 至 -25°C
复溶的储备溶液在-15至-25°C下稳定。
注：适当分装保存并避免反复冻融！

重悬

复溶于水中（100mg/mL）后续稀释采用PBS(磷酸缓冲盐)(不含Ca2+/Mg2+)。
