Cell Counting Kit-8 (CCK-8)细胞计数试剂盒采用WST-8 (2-(2-甲氧基-4-硝苯基)-3-(4-硝苯基)-5-(2,4-二磺基苯)-2H-四唑单钠盐)实现快速检测，其原理是WST-8能在电子载体1-甲氧基PMS的作用下被还原成水溶性甲臜产物。CCK-8溶液可直接加入到细胞中，不需要与其它组分预混。WST-8被细胞内脱氢酶生物还原后生成的橙色甲臜染料能够溶解在组织培养基中，生成的甲臜量与活细胞数量成正比。由于CCK-8溶液非常稳定并且细胞毒性很小，所以可以进行长时间孵育，例如孵育24-48小时。
Cell Counting Kit-8可以对细胞增殖和细胞毒性实验中活细胞数目进行敏感的比色法测定。检测灵敏度高于任何其它四唑盐，如 MTT、XTT或MTS。
