一般描述
甜菜碱也称为三甲基甘氨酸或N,N,N三乙基乙酸铵,是具有三个甲基的甘氨酸类似物。这种成分高度兼容聚合酶链式反应(PCR)缓冲液混合物。甜菜碱是一种PCR增强试剂,广泛用于提高PCR产物产量和特异性——尤其是在PCR扩增高GC含量的靶标,或PCR扩增形成二级结构、导致产量不佳的靶标过程中。甜菜碱可促进DNA链分离,并管理DNA中GC和AT对之间的DNA熔化温度(Tm)差异。它通过平衡GC-和AT-碱基对的贡献来稳定ds DNA。甜菜碱广泛用于优化多重和“长链且精确”的聚合酶链式反应(LA-PCR)。据报道,向PCR混合物中添加1.0-1.7M甜菜碱水溶液,可减少碱基对组成对DNA链解链的依赖性。
