Biochem/physiol Actions

Brain derived neurotrophic factor (BDNF) belongs to a family of secreted proteins called neurotrophins that includes, nerve growth factor (NGF), neurotrophin-3 (NT3) and NT4/5. BDNF is expressed in the developing and the mature brain. The downstream pathways promoted by BDNF include PI3K, MAPK and JAK/STAT. The most important role of BDNF is the regulation of synaptic transmission and plasticity, protein synthesis and phosphorylation of local proteins.

During development, neural networks are established in a highly organized manner, which persists throughout life. Neurotrophins play crucial roles in the developing nervous system. Drug-resistance to chemotherapics in aggressive neuroblastoma (NB) is characterized by enhanced cell survival mediated by TrkB and its ligand, brain-derived neurotrophic factor (BDNF); thus reduction in BDNF levels represent a promising strategy to overcome drug-resistance, but how chemotherapics regulate BDNF
