Keratinocyte Growth Medium 2 is a serum-free medium developed for the cultivation of epidermal keratinocytes without feeder cells. The medium is optimized for primary human cells but can also be used for horse and porcine keratinocytes, as well as mouse and rat keratinocytes after adapting the CaCl2 concentration. This allows users to adjust the CaCl2 concentration individually. To ensure the maximal growth performance of normal human Keratinocytes, PromoCell recommends adding the entire 60 μlCaCl2 stock solution, which results in final 0.06 mM CaCl2 concentration. In case a higher CaCl2 concentration is required (e.g. differentiation experiments), a 0.5 M CaCl2-Solution can be ordered separately (C-34006).

Store the SupplementPack at -20°C in the dark immediately after arrival. If stored properly, it is stable until the expiry date stated on the label. After adding the supplements to the Basal Medium, the shelf life of the complete medium is 6 weeks at 4 to 8°C. Do not freeze the complete medium. For use, pre-warm only an aliquot of the complete medium and keep the remaining medium refrigerated at 4 to 8°C.

Each lot is checked for growth promoting activity, adherence rate, and typical morphology of the tested keratinocytes. Approved inhouse lots of media are used as a reference. In addition, all lots of media have been tested for the absence of microbial contaminants (fungi, bacteria, mycoplasma).
