General description

氯喹是喹诺酮家族的一员，是一种弱的嵌入剂。氯喹用于治疗阿米巴病、类风湿性关节炎、盘状和系统性红斑狼疮。
氯喹能有效消灭各个发育阶段的红细胞型疟原虫，但对孢子虫没有影响。它还是一种抗生素。此外，浓度为 200 毫克/毫升（PBS，pH 值为 5.0）的氯喹可用于解离抗原-抗体复合物，而不会使红细胞抗原变性。最新研究表明，氯喹可作为一种抗肿瘤药物，与化疗和放疗一起用于癌症治疗。氯喹的抗疟作用是通过抑制血红素聚合成造血素来实现的，造血素是疟原虫的食物来源。氯喹与药物--安息香酸形成复合物，封住聚合链，阻止其进一步聚合。因此，游离血红素会积聚在食物泡中，对寄生虫产生毒性作用。此外，氯喹还能与T辅助细胞17上的转录因子结合，抑制其分化，从而起到抗自身免疫治疗的作用。二磷酸氯喹（CQ）经常被用作自噬途径的抑制剂。二磷酸氯喹和水杨甙联合使用，可通过调节线粒体功能障碍和自噬作用启动人肝细胞的凋亡。
