General description
产品为澄清至浑浊的无色溶液,以1 mg/ml 的浓度溶于含有2 μl 乙酸(或 0.1 N 乙酸)的水中时存在少量不溶物。通过沉降或离心可去除此不溶物。
所有胶原蛋白分子均由以三螺旋构象排列的三条肽链组成,其一级结构主要以三个氨基酸作为重复基序,即每隔两个氨基酸是一个甘氨酸残基,其前面的两个位置通常为脯氨酸或4-羟基脯氨酸。I型胶原不同于其他胶原蛋白的方面为其低赖氨酸羟基化和低碳水化合物构成。Col1a1(胶原蛋白,I型,α1)或胶原蛋白是一种主要的人体结构蛋白,它以纤维的形式聚集。这是一种很长很薄的蛋白质,是人体中发现的最丰富的蛋白质。它是由三条左旋多肽链组成的超螺旋右螺旋结构。这些链由~1040个氨基酸组成,基本上是三个氨基酸-(Gly-X-Y)n的重复序列。Gly是甘氨酸,X和Y可以是任何氨基酸,但人体中通常分别是脯氨酸和羟脯氨酸。

Biochem/physiol Actions
胶原蛋白是结缔组织的基本成分。一个中国家庭的研究表明COL1A1(胶原酶I型)突变与I型成骨不全有关。胶原蛋白和软骨下骨转换能力下调有关,可作为骨关节炎的关节间隙狭窄和骨刺状态的标志物。鼠尾胶原蛋白用于在组织培养板上铺上薄层包被,促进锚定依赖性细胞附着,推荐使用浓度为 6-10 μg/cm2。本品不能用于制作3-D凝胶。I型胶原常作为细胞培养的附着基质,与成肌细胞、脊髓神经节、肝细胞、胚胎肺、心脏外植体、成纤维细胞、内皮细胞、胰岛细胞一起在I型胶原的膜或凝胶上成功培养。I型胶原也可用于特发性肺纤维化(IPF)的研究,研究内质网应激IPF对肺成纤维细胞的影响。酸性溶液中的胶原蛋白可以产生三维支架,用于生物工程和细胞培养。
