应用

IV 型胶原在血管生成、神经疾病和转移中起关键作用。来自人胎盘的胶原蛋白已用于评估奎宁对气道上皮细胞的生物电效应、研究工程化慢病毒裂解肽的选择性毒性以及用于颗粒聚集测定以快速检测金黄色葡萄球菌（ Staphylococcus aureus）上的纤连蛋白、纤维蛋白原和胶原蛋白受体。在表征人肺肿瘤相关抗原过程中，它还用于镁依赖性胶原结合分析。

生化/生理作用

在发育期间，胶原蛋白 IV 广泛分布在 BM 中。在成熟过程中，该网络以显著的组织特定方式被部分取代，从而定义 BM 结构和功能。已显示胶原蛋白 IV 结合血小板、肝细胞、角质形成细胞、内皮细胞、肾小球膜细胞、胰腺细胞和一些肿瘤细胞。

自身免疫性疾病肺出血肾炎综合征中的组织损伤是由靶向胶原蛋白 IV α3 链的致病性自身抗体引起的。COL4A5 的突变与 Alport 综合征有关。

组分

所有胶原分子由以三螺旋构象排列的三条多肽链组成，其主要结构主要是在每三个位置具有甘氨酸的重复基序，并且脯氨酸或 4-羟基脯氨酸经常在甘氨酸残基之前。IV 型胶原仅在基底膜中发生，并含有多达六个遗传上不同的 α-链。

注意

该产品应在-20°C下干燥保存，在此条件下可保持活性3年。

制备说明

该粉末可在无菌条件下复溶。5 M 乙酸、PBS 或 1 mg/mL 水。PBS 溶液在-20℃ 下可稳定保存至少一年。

分析说明

SDS聚丙烯酰胺凝胶电泳在和基底膜胶原蛋白相同的还原条件下测试，得到三条主要条带。

其他说明

胶原蛋白被分为许多种结构和遗传性不同的类型。 我们使用 Bornstein和Traub提出的系统命名法。 小心不要混淆Sigma命名系统和通用的胶原蛋白分类类型。
