General description

I型胶原蛋白是皮肤、骨骼、肌腱和其他纤维结缔组织的组成部分。它由重复的三联体氨基酸甘氨酸、脯氨酸和羟脯氨酸组成。胶原蛋白是具有三个多肽链的左手螺旋结构。它是细胞外基质的组成部分，在牛中其有近28种类型。

Application

小牛皮肤的胶原蛋白已被用于：
  • 免疫荧光研究中载玻片的预先铺制
  • 在神经元培养中，作为细胞粘附因子和聚偏二氟乙烯-三氟乙烯（P（VDF-TrFE））膜的修饰
  • 在鼠胚成纤维细胞培养中用于培养皿的涂层

该产品旨在在组织培养板上产生薄层涂层，以促进贴壁依赖性细胞的附着，推荐用量为 6-10 μg/cm2。 本品不能用于制作3-D凝胶。 I型胶原常作为细胞培养的附着基质，与成肌细胞、脊髓神经节、肝细胞、胚胎肺、心脏外植体、成纤维细胞、内皮细胞、胰岛细胞一起在I型胶原的膜或凝胶上成功培养。I型胶原也可用于特发性肺纤维化(IPF)的研究，研究内质网应激IPF对肺成纤维细胞的影响。 酸性溶液中的胶原蛋白可以产生三维支架，用于生物工程和细胞培养。

Biochem/physiol Actions

热变性的I型胶原蛋白可导致三重螺旋分裂成随机卷曲。胶原基因的突变与多种牛疾病有关。它可用于食品和化妆品领域中。胶原蛋白在组织工程中可用作生物材料和组织支架。I型胶原纤维组装对于生理过程和细胞信号转导来说至关重要。

Preparation Note

Other Notes

胶原蛋白被分为许多种结构和遗传性不同的类型。我们使用Bornstein和Traub提出的命名法。不要混淆Sigma分类标识和公认的胶原蛋白分类类型。
