Contact inhibition and transformation studies

Derived from normal kidney and exhibits contact-inhibition. It is sensitive to viral or chemical transformation, including proteins such as SGF. Originated from the same mixed culture as NRK-52E but has distinct characteristics.

Rat kidney fibroblast

DMEM + 2mM Glutamine + 1% Non Essential Amino Acids (NEAA) + 5-10% Foetal Bovine Serum (FBS) or New Born Calf Serum (NBCS).

Split sub-confluent cultures (70-80%) 1:3 to 1:6 i.e. seeding at 2-4x10,000 cells/cm² using 0.25% trypsin or trypsin/EDTA; 5% CO₂; 37°C. The cells should be maintained sub-confluent, otherwise they will transform.
