Tumourigenicity studies

Derived from skin from the upper arm of an 18 year old Caucasian male with Gardner′s syndrome is an autosomal dominant condition which predisposes to carcinoma and multiple polyps of the colon. Cells exist in an initiated state rather than a normal or transformed state. Normal diploid karyotype.

Human Caucasian skin fibroblast Gardner′s syndrome

EMEM (HBSS) + 2mM Glutamine + 1% Non Essential Amino Acids (NEAA) + 10% Foetal Bovine Serum (FBS).

Split semi-confluent cultures 1:2 i.e. seeding at 4x10,000 cells/cm² using 0.25% trypsin; 5% CO₂; 37°C. Monolayer will form in a 25cm² flask in 1 week.
