Derived from a muscle tissue culture of a foetal Swiss-Webster mouse. Tumorigenic and only slightly sensitive to acetylcholine. When confluent cultures form short multinucleated myotubes.
Mouse Swiss-Webster myoblast
Preparation Note
DMEM + 2mM Glutamine + 10% Horse Serum (HS) + 10% Foetal Bovine Serum (FBS).
Split sub-confluent cultures (70-80%) 1:4 to 1:10 i.e. seeding at 1-2x10,000 cells/cm2 using 0.25% trypsin in Ca2+ + Mg2+-free PBS; 5% CO2; 37°C. Myotubes are formed at confluency, this is assisted by reducing both sera to 4%.
