Rat embryonic fibroblasts RSV-33 transformed
The R74 line was derived by co-cultivation of 18 day-old Lewis rat embryonic fibroblasts with cells of a chicken positive for Rous sarcoma virus-33 (RSV-33). This cell population is clearly distinguishable from RT1 and R9 as it is characterised by the presence of fibroblastoid cells that tend to round up and form clusters of refractile cells. R74 is also tumourigenic in chickens and rats.
Preparation Note
EMEM (EBSS) + 2mM Glutamine + 1% Non Essential Amino Acids (NEAA) + 5% Foetal Bovine Serum (FBS).
Split sub-confluent cultures (70-80%) 1:2 to 1:4 i.e. seeding at 2-4x10,000 cells/cm2 using 0.25% trypsin/EDTA; 5% CO2; 37°C.
