BALB/c mouse spleen x Mouse P3X63Ag8.653
The hybridoma cell line CAR-XP10, also known as XP10, was produced by fusing P3X63Ag8.653 mouse myeloma cells with spleen cells from a BALB/c mouse immunised six times with 20μg human thioredoxin (TRX) at monthly intervals. Human thioredoxin was cross-linked to Keyhole Limpet Haemocyanin for this purpose. The secreted antibodies react with monomeric and polymeric forms of human TRX and show no cross-reactivity to bacterial TRX or human glutaredoxin. CAR-XP10 has been used as second antibody in chemiluminescent detection assays for full human TRX. CAR-2/3F4 (Sigma Catalogue number. 97011602) can be applied as capture antibody.
Preparation Note
Maintain cultures between 3-9 x 100,000 cells / ml; 5% CO2; 37°C; start culture on mouse macrophage feeder layer
RPMI 1640 + 2mM glutamine + 1mM Sodium Pyruvate (NaP) + 15% Foetal Bovine Serum (FBS)
