Anti-Mouse CD19 (1D3) ColorWheel^® Dye-Ready mAb

ColorWheel^® technology is a novel and proprietary method of creating your own antibody and dye combinations for use in flow cytometry. By incubating any ColorWheel^® antibody with any ColorWheel^® dye, researchers can quickly and simply produce primary conjugated antibodies for use in single color or multicolor flow cytometry analysis. Each ColorWheel^® antibody and dye is lyophilized for long long-term storage, allowing for simplicity and flexibility without compromise. ColorWheel^® technology requires both ColorWheel^® antibody and ColorWheel^® dye for Flow Cytometry application.

B-lymphocyte antigen CD19 (UniProt: P25918; also known as Differentiation antigen CD19, CD19) is encoded by the Cd19 gene (Gene ID: 12478) in murine species. CD19 is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-16) and a propeptide (aa 17,18), which are subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 19-287), a transmembrane domain (aa 288-311), and a cytoplasmic domain (aa 312-547). It also contains two Ig-like C2-type domains (aa 20-113 and 171-271). CD19 is expressed in B cells in spleen, bone marrow, thymus and lymph nodes. It is also detected in peripheral blood lymphocytes. It associates with CD21 and CD81 to function as a co-receptor for the B cell receptor and reduces the threshold for activation of downstream signaling pathways and for triggering B cell responses to antigens. It is shown to be essential for normal B cell differentiation and proliferation in response to antigen challenges. Activation of CD19 is dependent on sequential phosphorylation of tyrosine residues 522 and 402 in the cytoplasmic domain by Lyn. It also undergoes phosphorylation at tyrosine 493, but this occurs more slowly and is transient and returns rapidly to basal levels. (Ref.: Ishiura, N., et al (2010). Eur. J. Quality Control Testing:

Evaluated by Flow Cytometry in mouse splenocytes.

Staining of one million mouse splenocytes was performed using 5 μL of a 1:1 mixture of Cat. No. CWA-1048, Anti-Mouse CD19 (1D3) ColorWheel^® Dye-Ready mAb and Cat. No. CWD-PE ColorWheel^® Antibody-Ready Phycoerythrin (PE) Dye or an equivalent amount of PE-conjugated Rat IgG2a isotype control.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user

Clone 1D3 is a rat monoclonal antibody that detects murine CD19. It targets an epitope within the extracellular domain.

Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.

1.0 mg/mL after reconstitution at 25 μL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 μL of PBS. Protect from light.
