General description

纤连蛋白（FN）/FN1（纤连蛋白 1）是由肝星状细胞（HSC）产生的多功能糖蛋白。它是细胞外基质（ECM）的组成部分。FN1 存在于细胞膜和细胞质中。它在体内由成纤维细胞、内皮细胞、巨噬细胞和肝细胞合成和释放。

Application

纤连蛋白大鼠血浆已用于诱导分化、粘附于细胞外基质、神经分化和原代神经元腹侧中脑培养物。

Biochem/physiol Actions

纤连蛋白（FN）/FN1（纤连蛋白 1）在细胞周期进程、细胞粘附和增殖以及纤维化进程中起主要作用。它对于胶原蛋白基质组装至关重要。FN 的高表达在肝纤维化过程中具有重要作用。FN1 有助于支持其他基质蛋白。

Preparation Note

冻干品，来自 pH 7.5，0.05M Tris 缓冲盐水。它在水中可溶于 1mg/mL，并应在 37℃ 下溶解至少 30 分钟。可能残留少量未溶解的物质，但不影响性能。蛋白质同质性经免疫电泳评估，呈现单弧线，经过内毒素测试，并且经测试源材料为 HIV、乙型肝炎和丙型肝炎阴性。

在涂层培养表面，纤维连接蛋白应在无菌平衡无菌盐溶液中稀释，并涂覆最小体积。然后将表面在室温下空气干燥 45 分钟，在 2-8℃ 下可储存 2-4 周。

Analysis Note

与纤维连接蛋白抗体相互作用，经 IEP（免疫电泳）分析呈现单弧线。

Other Notes

纤连蛋白是在细胞表面和血浆中发现的粘附糖蛋白。
