General description

纤连蛋白（FN）是一种多功能糖蛋白，相应的基因定位于人染色体2q34-36。该基因有75kb长并含有50个外显子。纤连蛋白结构中包含多个重复肽序列，这些序列在本质上是同源的，并在两个亚基上形成线性排列的功能单元。它存在有两种主要的异构体，一个可溶而另一种不可溶。前者存在与血浆中，而不可溶形式则存在于组织和软骨的细胞外基质（ECM）中。因在不同的区域存在可变剪切，故存在约20种不同的纤连蛋白形式，其中有两个III型外显子的被称为Extra Domains A和B。

细胞接种和神经元分化（作为CBSC（脐带血干细胞）重悬的扩增培养基的组分之一，并用于分化培养基）

Biochem/physiol Actions

纤连蛋白（FN）在多种生物学功能中起着必不可少的作用，如血管生成、细胞迁移、细胞-细胞粘附、细胞-基膜粘附、血栓稳定、胚胎发生、分化、伤口愈合以及病原体与哺乳细胞和胞外基质的结合。对中国汉族人群的研究表明该基因的突变可能与膝关节骨性关节炎（OA）的易感性存在关联。FN的的剪切体extra domain-B（EDB）是肿瘤血管生成的标志，并随着基质硬度的增加，内皮细胞而产生的这种异构体也会增加。在结直肠癌（CRC）中，EDA与肿瘤转移、血管生成和肿瘤生成存在关联，并可促进上皮-间充质转化。

Preparation Note

该产品是由pH 7.5的0.05 M Tris盐缓冲液冻干而得。以1 mg/mL浓度溶于水，在37°C下溶解至少30分钟。少量未溶解的物质可能会残留，但不会影响性能。通过免疫电泳评价蛋白的均匀性，检测内毒素，源材料HIV、乙肝、丙肝和梅毒检测均为阴性。在涂覆培养表面时，纤连蛋白应使用无菌平衡盐溶液进行稀释，并以最小体积进行涂覆。表面随后应在室温下空气干燥45分钟，并可在2-8°C储存2-4周。
