β-葡糖醛酸糖苷酶(GUSB)是一种溶酶体酶,存在于动物、植物和细菌中。这种糖苷外切酶在脾脏、肝脏和肾脏中的活性很高。牛肝β葡糖醛酸糖苷酶(BLG)是同源四聚体,分子量为290 kDa。

应用

β-葡萄糖醛酸酶在GUS测定中可用作报告基因以监测基因的表达。
β来自牛肝的-葡糖醛酸苷酶已被用于β-葡糖醛酸苷酶抑制试验,用于研究苦味叶下珠提取物和分离的活性成分对β-葡糖醛酸苷酶的抑制作用。它还被用于进行脱葡萄糖醛酸化。

生化/生理作用

β-葡糖醛酸糖苷酶(GUSB)缺乏会导致VII型粘多糖贮积症(MPSVII),从而导致溶酶体在大脑中的存储。类固醇激素的葡糖醛酸糖苷酶在组织中的整合β可以在GUSB的帮助下完成。
β-葡萄糖醛酸酶可催化复合碳水化合物的分解。在人体中,它可将结合胆红素转化为非结合的形式,使胆红素利于重吸收。

单位定义

在37℃,pH 5.0(30分钟检测)条件下,一个Sigma或改良的Fishman单位可每小时从酚酞葡糖苷酸中释放1.0 μg酚酞。
