Biochem/physiol Actions

Small heat shock proteins (HSPs) play a vital role in stress tolerance. They act as molecular chaperones in vitro. Murine Hsp25 averts aggregation of citrate synthase and α-glucosidase. It also inhibits actin polymerization and the amyloid fibril formation of Aβ1-42. Mutations in human sHsps result in myopathies, neuropathies and cataract. The small HSPs have also been implicated in phenomena of acquired thermotolerance and neoplasia.
The antibody also reacts with a dimer of the HSP 25 (approx. 58 kDa) appearing in preparations stored for prolonged periods prior to electrophoretic analysis. A major proteolytic fragment of 22 kDa may also be detected. The antibody suppresses the inhibition of actin elongation induced by the 25 kDa IAP. The antibody displays a punctate or fine fibrillary pattern of labeling throughout the cytoplasm in heat-shocked cells and co-localization with mitochondria. Reactivity has been observed with HSP 25 of various organs.

General description

Monoclonal anti-Heat Shock Protein 25 (mouse IgG1) isotype) is derived from the IAP-28 hybridoma produced by the fusion of mouse myeloma cells and splenocytes from BALB/c mice immunized with partially purified inhibitor of actin polymerization (IAP) protein from turkey gizzard smooth muscle. Heat shock (stress) proteins (HSPs) are a group of polypeptides synthesized in response to a wide variety of environmental factors, including biotic and abiotic stressors. These proteins are produced by prokaryotic and eukaryotic cells, and HSPs are among the most conserved molecules in phylogeny. Heat shock protein 25 is encoded by the gene mapped to human chromosome 7q11.23. The encoded protein belongs to the class of small HSPs. Small HSPs aggregate to form characteristic ring-shaped structures called heat shock granules. Small HSPs contains a-crystallin domain” (ACD) C-terminal domain, N-terminal domain and C-terminal extension (CTE).

Preparation Note

For continuous use, store at 2-8 °C. For extended storage, the solution may be frozen in working aliquots. Repeated freezing and thawing is not recommended. Storage in "frost-free" freezers is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use.
