General description

乙酰化&磷酸组蛋白H3 (Ac-Lys9, pSer10) 单克隆抗体（小鼠IgG2a同型）来源于小鼠骨髓瘤细胞 (NS1) 与用合成的乙酰化、磷酸化组蛋白H3多肽免疫的BALB/c小鼠脾细胞融合产生的APH3-64杂交瘤。组蛋白H3是核小体DNA复合体的组蛋白八聚体之一。它的N端尾段富含赖氨酸。

Immunogen

合成型乙酰化和磷酸化组蛋白H3肽（氨基酸7-20，Ac-Lys9，pSer10），对应于人组蛋白H3的N端。 该序列在许多物种中是相同的。

Biochem/physiol Actions

乙酰化&磷酸组蛋白H3（Ac-Lys，pSer）单抗特异性识别同时具有乙酰化赖氨酸和磷酸化丝氨酸的人组蛋白。
组蛋白乙酰转移酶 (HAT) 由组蛋白乙酰转移酶 (Gcn5p)、p300/CREB结合蛋白相关因子 (P/CAF)、E1A结合蛋白p300/CREB结合蛋白 (p300/CBP) 和转录起始因子TFIID 250 kDa亚基 (TAFII250) 组成，介导与转录激活有关的组蛋白N端结构域中赖氨酸的乙酰化。该修饰会引起核小体改构，使其更易与转录复合物结合。大多数物种中，组蛋白H3的主要乙酰化位点为赖氨酸9、14、18和23。在某些生物中，赖氨酸9的乙酰化在组蛋白沉积和染色质组装中起主导作用。组蛋白H3 Ser10位点的磷酸化与有丝分裂和减数分裂期间的染色体浓缩关系密切。丝氨酸位点的磷酸化与c-jun、c-fos和c-myc等立早癌基因的诱导有关。蛋白激酶A (PKA)、90kDa核糖体S6激酶-2 (Rsk-2) 和丝裂原和应力活化激酶 (Msk-1) 是组蛋白H3磷酸化的必需因子。ERK和p38通路会激活Msk-1以便磷酸化组蛋白H3。乙酰化和磷酸化组蛋白H3的单克隆抗体是研究染色质重塑和基因调节的重要工具。

Physical form

0.01 M 磷酸盐缓冲液，pH 7.4，含有 15 mM 叠氮化钠。

Preparation Note

若需连续使用，可在2-8°C下保存最长一个月。为了长期储存，请将样品等分，并在-20 °C下冷冻保存。不建议反复冻融。也不建议在无霜冰箱中储存。如果长期储存时出现轻微浑浊，请在使用前通过离心澄清溶液。若工作稀释样品在12小时内未使用完，则应丢弃。
