应用

潮霉素B抗生素可用于以下细胞的抗潮性作用：
  • 160 μg/ml，用于经质粒混合物转染的293T细胞。
  • 150 g/ml，用于MCF-7ALOX12细胞。
  • MSCV荧光素酶PGK-hygro质粒。
  • STAT3报告基因HeLa稳定细胞系（通过受STAT3启动子控制的荧光素酶基因稳定转染），维持在补充有10％FBS的DMEM中。
潮霉素B是一种氨基糖苷类抗生素，分离子从吸水链霉菌。 它已被用于研究70S核糖体易位和mRNA模板误读水平的蛋白质合成；作为一种抗病毒剂，通过选择性穿透被病毒感染渗透的细胞并抑制翻译；以及作为潮霉素抗性基因转化细胞的选择剂。 建议以100-800 μg/mL的浓度用作选择剂，其中对于原核生物使用100 μg/mL，对于低等真核生物使用200 μg/mL，对于高等真核生物使用150-400 μg/mL。

生化/生理作用

作用机制：本品通过诱导原核生物中m-RNA模板的误读，抑制蛋白质合成，具有抑制翻译的作用。

抗菌谱：潮霉素B对细菌、真菌和高等真核细胞有抑制作用。

制备说明

该产物通过离子交换色谱法进行纯化。 潮霉素B可以>50 mg/mL的浓度溶于水，也可溶于甲醇或乙醇。 溶液应通过过滤而非高压灭菌进行灭菌。 另外，据报道潮霉素B溶液在冷冻时会失去活性，并且其在冷藏可保持稳定，因此应避免冷冻。

免责声明

潮霉素B产品应在2-8°C的温度下保存，其干燥固定形式可在2-8°C的温度下保存至少五年。其在37°C下可稳定30天。 （对于溶液）如果在2-8°C下保存，该溶液可在两年内保持稳定。
