应用

IV 型胶原在血管生成、神经疾病和转移中起关键作用。来自人胎盘的胶原蛋白已用于评估奎宁对气道上皮细胞的生物电效应、研究工程化慢病毒裂解肽的选择性毒性以及用于颗粒聚集测定以快速检测金黄色葡萄球菌（ Staphylococcus aureus）上的纤连蛋白、纤维蛋白原和胶原蛋白受体。在表征人肺肿瘤相关抗原过程中，它还用于镁依赖性胶原结合分析。

生化/生理作用

在发育期间，胶原蛋白 IV 广泛分布在 BM 中。在成熟过程中，该网络以显著的组织特定方式被部分取代，从而定义 BM 结构和功能。已显示胶原蛋白 IV 结合血小板、肝细胞、角质形成细胞、内皮细胞、肾小球膜细胞、胰腺细胞和一些肿瘤细胞。


自身免疫性疾病肺出血肾炎综合征中的组织损伤是由靶向胶原蛋白 IV α3 链的致病性自身抗体引起的。COL4A5 的突变与 Alport 综合征有关。

组分

所有胶原分子由以三螺旋构象排列的三条多肽链组成，其主要结构主要是在每三个位置具有甘氨酸的重复基序，并且脯氨酸或 4-羟基脯氨酸经常在甘氨酸残基之前。IV 型胶原仅在基底膜中发生，并含有多达六个遗传上不同的 α-链。

将本产品储存在 -20°C。在这些条件下，它可保持活性 2 年。

制备说明

该产品以 0.5M 乙酸溶液形式提供。它可用于涂覆组织培养表面，促进细胞附着。这些应用中的用量取决于细胞类型、塑料器皿和孵育时间。推荐量为 0.5-5 μg/cm2。它通过胃蛋白酶提取和色谱纯化制备。
