General description
SCAMP2 (secretory carrier membrane protein 2) belongs to the SCAMP family of highly conserved integral membrane proteins, which are involved in cell surface recycling pathways). SCAMP2 is one of the two isoforms of SCAMP, the other being SCAMP1, and has a molecular weight of 39kDa. It has a wide range of tissue expression. It is a transmembrane protein, which contains a cytoplasmic N-terminal, cytoplasmic C-terminal tail and four transmembrane regions. It contains NHE5 (Na+/H+ exchanger)-binding sites in both its N- and C-termini. It also contains the characteristic E-peptide of SCAMP proteins, within the second and third transmembrane helices. This protein resides in endosomes, other recycling organelles, as well as plasma membrane.

Biochem/physiol Actions
SCAMP2 (secretory carrier membrane protein 2) is involved in Ca2+-dependent granule exocytosis, and is a part of the later stages of membrane fusion. It controls the formation of membrane fusion pores, by sequestering phosphatidylinositol 4,5-bisphosphate (PIP2) within membranes. It interacts with PIP2 via a hydrophobic sequence CWYRPIYKAFR, present in its structure. It interacts with serotonin and dopamine transporters, and controls their subcellular distribution and their surface expression. It interacts with NHE5, which is a Na+/H+ exchanger highly expressed in brain, through its cytoplasmic N- and C-termini. This way, it regulates the movement of NHE5 through recycling endosomes, and its cell surface targeting, in the presence of Arf6 (ADP-ribosylation factor 6). The E-peptide of SCAMP2 protein is a potent inhibitor of exocytosis during the later stages.
