The protein is expressed on the epithelial brush border in the small intestine, colon, gall bladder, renal proximal tubule, thick ascending limb of the loop of Henle in the kidney and respiratory center. The gene is located on human chromosome 5p15.3.

Inhibition of SLC11A1 is linked to diarrheal diseases and sudden infant death syndrome. The protein plays a pivotal role in the pathogenesis of cholesterol gallstone disease (CGD).
