General description
MADD (MAP kinase-activating death domain protein) is located on human chromosome 11p11 and it consists of 36 exons. It is also called as DENN or Rab3GEP. The IG20 (insulinoma glucagonoma clone 20) gene codes for MADD.

Biochem/physiol Actions
MADD (MAP kinase-activating death domain protein) prevents apoptosis and induces human lung adenocarcinoma. It has the ability to bind to the death domain of TNFR1 (tumor necrosis factor receptor 1). MADD can activate TNF-α gene expression and also has the capability to promote the JNK (c-Jun NH2-terminal kinase) pathway. It modulates the recycling of Rab3 small G proteins and plays a major role in Ca2+-dependent neurotransmitter release and exocytosis. This protein also participates in inhibiting the apoptosis of neuronal cells.
