一般描述

无血清培养可避免血清带来的成分差异和污染。无血清配方将通常由血清提供的因子替换为纯化形式，适合许多体外生长和分化研究。这些纯化因子包括胰岛素、转铁蛋白、硒、丙酮酸和乙醇胺。其他补充成分并不一定，具体取决于研究的细胞类型和所用的基础培养基。ITS补充剂是胰岛素、人转铁蛋白（部分铁饱和）和亚硒酸钠的混合物。它是一种通用的细胞补充剂，设计用于含有丙酮酸钠的非复合型培养基（如MEM、RPMI-1640）及复合型培养基（如Ham′s F-12、DME/F-12、MEM）。

应用

ITS+1液体培养基补充剂(100×)已用作高糖杜氏改良Eagle培养基(DMEM)的补充剂，用于小鼠间充质干细胞(mMSC)向软骨细胞的分化。还用作基础分化培养基的补充剂，用于在无血清条件下使人多能诱导干细胞(hiPSC)分化为成骨细胞。
胰岛素 - 转铁蛋白 - 亚硒酸钠，亚油酸 - BSA已用于无血清细胞培养应用.ITS + 1液体培养基补充剂（100×）已用于培养细胞系和用于制备永生化培养基，以用于生产来自′永生鼠′胚胎的细胞系。

制备说明

使用不含酚红的Earle′平衡盐溶液（EBSS）制备。

其他说明

含 1.0 mg/mL 牛胰岛素、0.55 mg/mL 人转铁蛋白（基本上无铁）、0.5μg/ml 亚硒酸钠、50 mg/mL 牛血清白蛋白和 470μg/ml 亚油酸（100x 浓度）。
