一般描述
MES是针对生物应用而开发的“良好”缓冲液之一。该产品经过痕量离子测试。采用氢氧化钠滴定游离酸至目标pH (pKa=±-1),可以制备含MES游离酸的缓冲液。另外,将MES 游离酸和MES钠盐的等摩尔溶液混合起来,可以得到目标pH。采用储备溶液制备给定pH缓冲液所需的标准品混合表已经公布。
应用
MES钠盐已被用于:
- 在含基因组DNA的琼脂糖块融化的过程中使DNA纤维达到目标密度和拉伸性
- 平衡均质机的样品室,并防止样品在均质之前水解
制备说明
MES 钠溶于水,50毫升水可溶解25克形成无色透明溶液(大约2.3M)。根据浓度不同,溶液的pH应当在9和10之间。溶液在2-8°C 下应当可稳定储存数月。灭菌应通过 0.2μm过滤器过滤完成。不建议用高压蒸汽灭菌。如果缓冲液必须不含核酸酶,则最好的做法是先对水进行处理,然后在高压蒸汽灭菌后添加缓冲液固体。MES溶液在高压蒸汽灭菌时会变成黄色(尽管pH没有出现可测量的变化)。黄色分解产物是未知的物质。
