一般描述

谷氨酸脱氢酶 (GDH) 是一种线粒体酶,催化谷氨酸可逆性氧化脱氨生成 α-酮戊二酸,作为合成代谢和分解代谢途径之间的关键链接。在哺乳动物中,GDH受变构调节,在肝脏、肾脏、大脑和胰腺中具有较高的活性。血清中的 GDH 活性可用于区分肝脏炎症所致的肝脏疾病(未显示血清 GDH 活性升高)和导致肝细胞坏死的疾病(导致血清 GDH 升高)。

原理

通过偶联酶试验可测定 GDH 活性,其中谷氨酸被 GDH 消耗,生成 NADH,NADH 与探针反应生成与 GDH 活性成比例的比色 (450 nm) 产物。一个单位的GDH 在37℃下pH 7.6 时 每分钟将产生1.0μ摩尔NADH的酶量。
