一般描述
神经氨酸酶,也称为唾液酸酶,是一种水解多糖链上末端唾液酸残基的酶。它主要在细菌和病毒等微生物中表达。据信神经氨酸酶切割唾液酸残基在流感病毒感染中起多种作用。人们认为它有助于粘膜衬里的渗透、靶细胞的入侵、子代病毒从感染细胞中的洗脱以及防止自聚集。因此,神经氨酸酶是流感药物开发的重要目标,用于测量神经氨酸酶活性的简单,直接和自动化的程序在研究和药物发现中得到了广泛的应用。

生化/生理作用
在该测定中,神经氨酸酶活性通过酶促测定法测定,其中神经氨酸酶释放的唾液酸产生与样品中神经氨酸酶活性成正比的比色(570 nm)/荧光(λex = 530/λem = 585 nm)产物。在37°C下的线性检测范围对于比色测定为0.1-10单位/L,对于荧光测定为0.01-2单位/L。
